Bioinformatics and functional analysis define four distinct groups of AlkB DNA-dioxygenases in bacteria

The iron(II)- and 2-oxoglutarate (2OG)-dependent dioxygenase AlkB from Escherichia coli (EcAlkB) repairs alkylation damage in DNA by direct reversal. EcAlkB substrates include methylated bases, such as 1-methyladenine (m(1)A) and 3-methylcytosine (m(3)C), as well as certain bulkier lesions, for example the exocyclic adduct 1,N(6)-ethenoadenine (epsilonA). EcAlkB is the only bacterial AlkB prote...

Estimation of Significance of Alkb and Alka Proteins in Dna Repair in Escherichia Coli Model

The paper concerns estimation of significance of differences of mutagenesis level between the wild-type strain (wt) and its derivatives which differ in DNA repair ability, namely alkA and alkB strain, devoided AlkA glycosylase and AlkB dioxygenase activity, respectively. The strains were analyzed for their ability to repair 1,N-ethenoadenine (εA) – chloroacetaldehyde adduct to DNA. The analysis...

Removal of N-Alkyl Modifications from N2-Alkylguanine and N4-Alkylcytosine in DNA by the Adaptive Response Protein AlkB

The AlkB enzyme is an Fe(II)- and α-ketoglutarate-dependent dioxygenase that repairs DNA alkyl lesions by a direct reversal of damage mechanism as part of the adaptive response in E. coli. The reported substrate scope of AlkB includes simple DNA alkyl adducts, such as 1-methyladenine, 3-methylcytosine, 3-ethylcytosine, 1-methylguanine, 3-methylthymine, and N(6)-methyladenine, as well as more co...

The Influence of Exocyclic Dna Adducts in Bacterial and Mammalian Genome Instability

Oxidative stress enhances lipid peroxidation (LPO) implicated in the promotion and progression of carcinogenesis. One of the major LPO products is trans-4-hydroxy-2-nonenal (HNE), may to react with guanosine and under peroxidizing conditions also with adenosine. Additionally the same effect may induce environmental carcinogens, e.g. vinyl chloride and its metabolite chloroacetaldehyde (CAA). Th...

Direct repair of 3,N(4)-ethenocytosine by the human ALKBH2 dioxygenase is blocked by the AAG/MPG glycosylase.

Exocyclic ethenobases are highly mutagenic DNA lesions strongly implicated in inflammation and vinyl chloride-induced carcinogenesis. While the alkyladenine DNA glycosylase, AAG (or MPG), binds the etheno lesions 1,N(6)-ethenoadenine (ɛA) and 3,N(4)-ethenocytosine (ɛC) with high affinity, only ɛA can be excised to initiate base excision repair. Here, we discover that the human AlkB homolog 2 (A...
